In this application, we will develop a nanoparticle formulation for the pancreatic tumor specific delivery of miR-145 and K-Ras inhibiting siRNA and investigate if this formulation effectively inhibits oncogenic mechanisms involved in tumor growth, metastasis, and drug resistance of PDAC. This project would establish that combination therapies could be delivered through our nanotechnology platform that selectively target PDAC tumor and macro/micro metastatic lesions with minimum toxicities upon systemic administration. Overall, this proposal is designed to enable in-depth pre-clinical studies along with the investigations on the mechanisms. This proposal has potential to develop a new therapeutic modality for pancreatic cancer especially those became resistant to frequently used chemotherapies such as gemcitabine.
